I have walked all of the route within the past few years.
There is more ascent then Watbig 1 & 2 but it avoids the tarmac and levelness of the Manifold Trail.
The River Manifold (many bends) is seasonal and in summer the flow is entirely underground here. It falls down holes just downstream of Wetton Mill and the resurgence is at Ilam Hall.
From Weag's Bridge (or via Grindon, see gargoyles at foot of church tower) you may want to detour upstream a bit to visit Thor's Cave, one of the wonders of the Peak District. There is a permissive path from the cave to Wetton which avoids losing any height. It is shown on OpenStreetMap but not the Ordnance Survey topographic and only partially on the Ordnance Survey standard view.
An alternative detour is downstream to Beeston Tor, across the stepping stones (dry in summer), and up a nice track and path to Wetton.
There is a pub in Wetton but check on opening. You can switch to Watbig 1 or 2 at Wetton.
There are some nice caves just downstream from where the route crosses the River Dove in Beresford Dale
